home services platforms illumina: gene expression
Illumina gene expression protocols feature a first- and second-strand reverse transcription step, followed by a single in vitro transcription (IVT) amplification that incorporates biotin-labelled nucleotides. Subsequent steps include array hybridisation, washing, blocking, and streptavadin-Cy3 staining. Fluorescence emission by Cy3 is quantitatively detected for downstream analysis. |
Service includes:
- Total RNA Quality Checked with Agilent BioAnalyzer ($10 fee is charged if RNA quality deemed too low quality to proceed with the experiment)
- Labelling of one RNA sample per array with streptavidin-cyanine-3
- Hybridisation/washing/scanning of arrays
- Feature quantification (BeadStudio)
- Quantification output files sent to customer (via Data Portal)
Sample Requirement:
We request 100ng of total RNA in nuclease-free water at a concentration greater than 50 ng/µL. If you have less than 100 ng total RNA, please contact us to review amplification options. The quality of the samples is checked upon arrival using the Agilent 2100 Bioanalyzer. Partially degraded samples or FFPE samples can be labelled using the DASL Assay (cDNA-mediated annealing, selection, extension, and ligation assay).
Array Types for Gene Expression:
Whole Genome BeadChips
